TB 500 is a synthetic variant of the naturally occurring peptide Thymosin Beta 4 (TB 500), which is found in nearly all human and animal cells. It is a pioneering drug candidate that facilitates the following processes: differentiation of endothelial (blood vessel) cells; angiogenesis (formation of new blood vessels from existing ones) in skin tissues; migration of keratinocytes; deposition of collagen; and reduction of inflammation.

Among the primary mechanisms of TB 500 is its ability to modulate Actin, a crucial protein that supports cell structure and movement. Of the myriad proteins found in cells, actin constitutes up to 10% of the total, thus playing a significant role in the cellular genetic makeup.

This powerful peptide belongs to a widespread family of 16 related molecules that exhibit a high degree of sequence and localization conservation across most tissues and circulating cells in the body. TB 500 not only interacts with actin but also inhibits actin polymerization and serves as the actin-sequestering agent in eukaryotic cells.

TB 500 was identified as a gene that increases its expression by four to six times during the initial stages of blood vessel formation and has been shown to encourage the proliferation of new blood cells from existing vessels. This peptide is found in wound fluid; when administered subcutaneously, it aids in wound healing, muscle development, and accelerates the recovery of muscle fibers and their cells.

Another crucial aspect of TB 500 is its facilitation of cell migration through a specific interaction with actin within the cell cytoskeleton. Research has shown that a central small amino acid long-actin binding domain possesses both blood cell reproduction and wound healing properties. These effects are realized by enhancing the migration of endothelial cells and keratinocytes, as well as increasing the production of enzymes that degrade the extracellular matrix.

Studies indicate that TB 500 is a powerful, naturally occurring wound repair factor with anti-inflammatory effects. Unlike other repair factors, such as growth factors, TB-500 promotes the migration of endothelial and keratinocyte cells. Additionally, it does not attach to the extracellular matrix and possesses a low molecular weight, allowing it to travel relatively long distances through various tissues.
